    The Effects of Temperature, Diet, and Other Factors on Development, Survivorship, and Oviposition of Aethina tumida (Coleoptera: Nitidulidae)

    Developmental rate and survivorship of small hive beetle, Aethina tumida Murray (Coleoptera: Nitidulidae), life stages were measured across different temperatures (21, 25, 28, 32 and 35 C) and diets, which included natural and artificial pollen, honey, and bee pupae. Temperature affected hatch success, time to hatching, and larval growth. Eggs hatched in 61 h at 21 C but in 22 h at 35 C. Larvae achieved peak weight in 8 d at 35 C but needed 17 d at 21 C. Diet had comparatively little effect on larval survivorship or maximum weight, although larvae fed only bee pupae had lower survivorship. Access to soil influenced pupation success. Duration of the life stage spent in the soil, during which pupation occurs, was also affected by temperature: adults emerged after 32.7 d at 21C but after only 14.8 d at 35 C, albeit with high mortality. Minimum temperature for development was estimated at 13.5 C for eggs, and 10.0 C for larvae and pupae. Temperature influenced adult longevity and oviposition: on a honey and pollen diet average adult lifespan was 92.8 d at 24 C but only 11.6 d at 35 C. Beetles lived longer at 28 C or lower but produced the most eggs per female, regardless of diet, at 32 C. Beetle density influenced fecundity: beetles kept at three pairs per vial laid 6.7 times more eggs per female than those kept as single pairs. Overall, beetles fared best at 28-32 C with mortality of all stages highest at 35 C

    Effects of bee density and sublethal imidacloprid exposure on cluster temperatures of caged honey bees

    International audienceAbstractSurvivorship, syrup consumption, and cluster temperatures of honey bees were kept in hoarding cages with different numbers of bees. Cages with either 50, 100, 150, or 200 bees each were monitored over 4–6 weeks in incubators with 12h/12h 30° C/15° C temperature cycles to induce clustering. Survivorship and syrup consumption rates per bee were not different among the bee density groups, but cluster temperatures were. Cluster temperatures ranged from 0.45°C above incubator temperature in the 50 bee cages to 4.05° C in the 200 bee cages over the 1st 7 days, with each additional bee adding on average 0.02° C to cluster temperature. In another set of experiments, cages were established with about 200 bees each, and imidacloprid added to the syrup at 0, 5, 20, and 100 ppb. Imidacloprid in the syrup did not affect bee survivorship but it did reduce syrup consumption per bee, with bees fed 100 ppb imidacloprid syrup consuming on average 631 mg per bee over 28 days while average consumption among the other groups ranged from 853 to 914 mg. Cluster temperature was affected by imidacloprid treatment: bees fed 5 ppb imidacloprid syrup had higher cluster temperatures over the 1st 10 days, 4.17° C above incubator temperature, than either bees fed 100 ppb syrup or control (2.35 and 3.19° C, respectively)

    Honey Bee Survival and Pathogen Prevalence: From the Perspective of Landscape and Exposure to Pesticides

    In order to study the in situ effects of the agricultural landscape and exposure to pesticides on honey bee health, sixteen honey bee colonies were placed in four different agricultural landscapes. Those landscapes were three agricultural areas with varying levels of agricultural intensity (AG areas) and one non-agricultural area (NAG area). Colonies were monitored for different pathogen prevalence and pesticide residues over a period of one year. RT-qPCR was used to study the prevalence of seven different honey bee viruses as well as Nosema sp. in colonies located in different agricultural systems with various intensities of soybean, corn, sorghum, and cotton production. Populations of the parasitic mite Varroa destructor were also extensively monitored. Comprehensive MS-LC pesticide residue analyses were performed on samples of wax, honey, foragers, winter bees, dead bees, and crop flowers for each apiary and location. A significantly higher level of varroa loads were recorded in colonies of the AG areas, but this at least partly correlated with increased colony size and did not necessarily result from exposure to pesticides. Infections of two viruses (deformed wing virus genotype a (DWVa) and acute bee paralysis virus (ABPV)) and Nosema sp. varied among the four studied locations. The urban location significantly elevated colony pathogen loads, while AG locations significantly benefited and increased the colony weight gain. Cotton and sorghum flowers contained high concentrations of insecticide including neonicotinoids, while soybean and corn had less pesticide residues. Several events of pesticide toxicity were recorded in the AG areas, and high concentrations of neonicotinoid insecticides were detected in dead bees

    Synthetic Ligands of Olfactory Binding Proteins Modulate Aggregation Response of Asian Citrus Psyllid in the Presence of Host-Plant Volatiles

    There is interest in using ligands of chemosensory binding proteins (CBP) to augment an insect’s responsiveness to chemosensory cues. We showed previously that combining a synthetic ligand of a CBP with limonene, a common citrus volatile, enhanced the probing response of Asian citrus psyllid (Diaphorinacitri). Here, we determined whether synthetic compounds, which were ligands of D. citri olfactory binding protein (OBP) DCSAP4, influenced the settling and aggregation levels of psyllids on young citrus shoots. The test ligands and Cmac scent were dispensed from a droplet of an emulsified wax product (SPLAT) placed on the bottom of each vial. The shoots were presented: (1) alone (shoot + blank SPLAT), (2) with a mixture of citrus volatiles (“Cmac scent”) (shoot + SPLAT with Cmac scent), or (3) with different concentrations of test ligands (shoot + SPLAT with test ligand at concentration 1, shoot + SPLAT with test ligand at concentration 2, etc.). Depending on the availability of test ligands, sprigs, and psyllids, each test included from two to four replicates of each treatment (i.e., shoot only, shoot + Cmac scent, shoot + test ligand at concentration 1, shoot + test ligand at concentration 2, etc.); only a single test ligand was presented in each test. For each test, 200 D. citri were released in the test area and the numbers of psyllids on each sprig were counted 24 h later. Sprigs with ≥7 psyllids were considered to be an aggregation. A total of seven ligands were tested individually. Four of the ligands (654, 717, 784, and 861) modulated psyllid settling and aggregation response, causing greater settling and aggregation to sprigs presented with the Cmac scent than to those sprigs with blank SPLAT. Presentation of one of the ligands (019) resulted in an opposite effect in which psyllid settling and aggregation levels were lower on sprigs with Cmac scent than on those with blank SPLAT. There were no differences in settling levels in the different treatment vials in the Ligand 905 experiment. In the Ligand 937 experiment, settling levels did not vary significantly between treatment vials although settling levels were relatively high in all treatment vials and there was a significant treatment effect. Increased settling and aggregation levels were largely not observed with in the vials with only the test ligands, and there was little effect of ligand concentration on psyllid response levels. This suggests that the test ligands themselves did not attract the psyllids but rather modulated the psyllid’s response to the Cmac scent. The results suggest that synthetic ligands of D. citri CBPs can be used to increase the effectiveness of citrus scent lures used to attract psyllids to monitoring traps and attract and kill devices

    Controlling anal incontinence in women by performing anal exercises with biofeedback or loperamide (CAPABLe) trial: Design and methods

    The goals of this trial are to determine the efficacy and safety of two treatments for women experiencing fecal incontinence. First, we aim to compare the use of loperamide to placebo and second, to compare the use of anal sphincter exercises with biofeedback to usual care. The primary outcome is the change from baseline in the St. Mark's (Vaizey) Score 24 weeks after treatment initiation. As a Pelvic Floor Disorders Network (PFDN) trial, subjects are enrolling from eight PFDN clinical centers across the United States. A centralized data coordinating center supervises data collection and analysis. These two first-line treatments for fecal incontinence are being investigated simultaneously using a two-by-two randomized factorial design: a medication intervention (loperamide versus placebo) and a pelvic floor strength and sensory training intervention (anal sphincter exercises with manometry-assisted biofeedback versus usual care using an educational pamphlet). Interventionists providing the anal sphincter exercise training with biofeedback have received standardized training and assessment. Symptom severity, diary, standardized anorectal manometry and health-related quality of life outcomes are assessed using validated instruments administered by researchers masked to randomized interventions. Cost effectiveness analyses will be performed using prospectively collected data on care costs and resource utilization. This article describes the rationale and design of this randomized trial, focusing on specific research concepts of interest to researchers in the field of female pelvic floor disorders and all other providers who care for patients with fecal incontinence

    High-throughput screening of monoclonal antibodies against plant cell wall glycans by hierarchical clustering of their carbohydrate microarray binding profiles

    Antibody-producing hybridoma cell lines were created following immunisation with a crude extract of cell wall polymers from the plant Arabidopsis thaliana. In order to rapidly screen the specificities of individual monoclonal antibodies (mAbs), their binding to microarrays containing 50 cell wall glycans immobilized on nitrocellulose was assessed. Hierarchical clustering of microarray binding profiles from newly produced mAbs, together with the profiles for mAbs with previously defined specificities allowed the rapid assignments of mAb binding to antigen classes. mAb specificities were further investigated using subsequent immunochemical and biochemical analyses and two novel mAbs are described in detail. mAb LM13 binds to an arabinanase-sensitive pectic epitope and mAb LM14, binds to an epitope occurring on arabinogalactan-proteins. Both mAbs display novel patterns of recognition of cell walls in plant materials

    Do Dispersing Monkeys Follow Kin? Evidence from Gray-cheeked Mangabeys (Lophocebus albigena)

    Among social vertebrates, immigrants may incur a substantial fitness cost when they attempt to join a new group. Dispersers could reduce that cost, or increase their probability of mating via coalition formation, by immigrating into groups containing first- or second-degree relatives. We here examine whether dispersing males tend to move into groups containing fathers or brothers in gray-cheeked mangabeys (Lophocebus albigena) in Kibale National Park, Uganda. We sampled blood from 21 subadult and adult male mangabeys in 7 social groups and genotyped them at 17 microsatellite loci. Twelve genotyped males dispersed to groups containing other genotyped adult males during the study; in only 1 case did the group contain a probable male relative. Contrary to the prediction that dispersing males would follow kin, relatively few adult male dyads were likely first- or second-degree relatives; opportunities for kin-biased dispersal by mangabeys appear to be rare. During 4 yr of observation, adult brothers shared a group only once, and for only 6 wk. Mean relatedness among adult males sharing a group was lower than that among males in different groups. Randomization tests indicate that closely related males share groups no more often than expected by chance, although these tests had limited power. We suggest that the demographic conditions that allow kin-biased dispersal to evolve do not occur in mangabeys, may be unusual among primates, and are worth further attention

    Interstellar-medium mapping in M82 through light echoes around supernova 2014J

    We present multiple-epoch measurements of the size and surface brightness of the light echoes from supernova (SN) 2014J in the nearby starburst galaxy M82. Hubble Space Telescope (HST) ACS/WFC images were taken ~277 and ~416 days after B-band maximum in the filters F475W, F606W, and F775W. Observations with HST WFC3/UVIS images at epochs ~216 and ~365 days are included for a more complete analysis. The images reveal the temporal evolution of at least two major light-echo components. The first one exhibits a filled ring structure with position-angle-dependent intensity. This radially extended, diffuse echo indicates the presence of an inhomogeneous interstellar dust cloud ranging from ~100 to ~500 pc in the foreground of the SN. The second echo component appears as an unresolved luminous quarter-circle arc centered on the SN. The wavelength dependence of scattering measured in different dust components suggests that the dust producing the luminous arc favors smaller grain sizes, while that causing the diffuse light echo may have sizes similar to those of the Milky Way dust. Smaller grains can produce an optical depth consistent with that along the supernova-Earth line of sight measured by previous studies around maximum light. Therefore, it is possible that the dust slab from which the luminous arc arises is also responsible for most of the extinction toward SN 2014J. The optical depths determined from the Milky Way-like dust in the scattering matters are lower than the optical depth produced by the dust slab
